[ Summary ] The α-Klotho gene encodes a single-pass transmembrane protein. In addition to the transmembrane form which is known as a coreceptor of FGF23, α-Klotho also exists in a soluble form. Soluble α-Klotho is detectable in serum, cerebrospinal fluids, and urine, and is considered to be a humoral factor. There have been some recent reports which have revealed that soluble α-Klotho is involved in mineral metabolism. A sandwich ELISA for soluble α-Klotho has been established. The measurement of soluble α-Klotho levels has revealed that there are some physiological relationships between values of soluble α-Klotho and other parameters involved in mineral homeostasis. In specific conditions such as perinatal period or in genetic disorders, the values of soluble α-Klotho exhibit indicative characteristics. From these findings, the measurement of soluble α-Klotho levels can be seen to be useful as a novel biomarker.
