[ Summary ] Glucocorticoids (GCs) are essential for the maintenance of homeostasis. However, GCs in excess doses cause various adverse effects. GCs have biphasic effects on osteoblast function. Although GCs at physiological concentrations promote differentiation of mesenchymal stromal cells into osteoblasts, GCs at pharmacological doses suppress osteoblast function by such mechanisms as suppression of Cbfal expression, reduction in production of various growth factors, promotion of osteoblast apoptosis and suppression of bone formation signals including Wnt, leading to glucocorticoid-induced osteoporosis.
