[ Summary ] The parathyroid gland function is regulated by extracellular calcium, vitamin D and phosphate in cooperation with the kidney, bone and intestine. PTH secretion is strictly regulated by the extracellular calcium concentration mediated by the G protein-coupled, calcium-sensing receptor (CaSR) situated on the chief cells of the parathyroid glands. Recent studies have been reported that some molecules including MAPK and a scaffold protein, Filamin, are involved in the CaSR signaling and that PTHrP production can be regulated through the CaSR as well. The mechanisms of PTH and PTHrP secretion/synthesis are discussed in this review.
