[ Summary ] If bone microdamage is repaired by bone remodeling, suppressed bone remodeling may increase microdamage accumulation and lead to increased bone fragility. In this series of studies, we evaluated the effects of suppressed bone remodeling caused by bisphosphonates on microdamage accumulation and biomechanical properties of dog bone. Amino-bisphosphonates increased or maintained bone strength due to increased bone mass without impairment of mineralization. Meanwhile, however, suppressed turnover for both intracortical and trabecular bone increased accumulation of microdamage and decreased some intrinsic material property of bone. High dose treatment of etidronate decreased mineralized bone mass and bone strength because of increased osteoid accumulation as a result of suppressed mineralization. However, microdamage accumulation was not prominent despite of complete suppression of bone remodeling. This indicates that osteoid accumulation prevent production of microdamage.
