[ Summary ] Cancer accounts for 1/3 of the deaths in Japan. It is thought that obesity is a factor leading to the development of cancer. Obesity linked to increased cancer risk factors, such as disturbed insulin levels, as well as IGF-1 and leptin imbalances have been analyzed in detail. Mechanisms related to carcinogenesis or cancer progression have also been investigated. The PI3K/Akt signaling pathway is activated by insulin, IGF-1 and leptin. This is due to obesity. These factors have been revealed as potential therapeutic targets. AMP kinase and mTOR are affected by obesity, and key molecules which control energy consumption and survival. Gut microbiota and physical activity may be involved in the initiation or progression of several forms of cancer.
