[ Summary ] In this decade, the amazing progress of molecular biological studies has revealed that colorectal carcinogenesis and the progression of cancer are associated with both numerical and structure disorders of human genes. By gaining new insight into colorectal carcinogenesis, various benefits in the management of colorectal cancer are now anticipated ; the possibilities include presymptomatic diagnosis of hereditary colorectal cancer, evaluation of malignant potentiality and the individualization of operative procedures, susceptibilities to anti cancer drugs and prediction of second primary cancers. With regard to this subject, we have described herein the recent advancement of molecular biological studies in colorectal carcinogenesis, focusing especially on two well known inherited colorectal cancers ; familial adenomatous polyposis (FAP) and hereditary nonpolyposis colorectal cancer (HNPCC).
