Osteoclast differentiation is tightly regulated by osteoblasts. Osteoblasts express two cytokines, M-CSF and RANKL, essential for osteoclast differentiation. Osteoclast precursors firstly differentiate into mononuclear osteoclasts in response to M-CSF and RANKL. NFATcl is a transcription factor, which is involved in the determination of osteoclast differentiation. NFATcl is activated by calcium oscillations. Calcium channels have an effect on calcium oscillations. Calcium channels can also play a role in osteoclast differentiation and function. The application of calcium channel inhibitors may be a new therapeutic strategy for the treatment of local bone resorption induced by inflammatory disease. |
