【 要旨 】 破骨細胞は造血幹細胞由来の単球/マクロファージ系に属する終末分化細胞であり,そのもっとも特徴的な細胞現象の一つに,単核の破骨細胞同士の細胞融合による多核化が挙げられる.われわれはこの破骨細胞の細胞融合を制御する分子を同定するため,細胞融合を起こした多核の破骨細胞と,細胞融合を起こさない単核のマクロファージとの間で発現分子のサブトラクションを行い,破骨細胞側に高発現する分子として dendritic cell specific transmembrane protein (DC-STAMP) を同定した.DC-STAMP欠損マウスを作製したところ,破骨細胞並びに異物巨細胞の細胞融合が完全に欠損することを見出した.

Title Regulation of osteoimmunology by DC-STAMP

[ Summary ] Osteoclasts are terminally differentiated cells derived from hematopoietic stem cells or monocyte / macrophage lineage cells. The most characteristic feature of osteoclasts is multinucleation by cell-cell fusion of mononuclear osteoclasts. In order to isolate the cell-cell fusion-inducing molecule in osteoclasts, we undertook DNA subtraction screening between multinuclear osteoclasts and mononuclear macrophages. It was found that DC-STAMP was highly expressed in osteoclasts compared with macrophages. To understand the role of DC-STAMP in vivo, DC-STAMP deficient mice were generated. DC-STAMP deficient mice show a complete lack of cell-cell fusion of osteoclasts and foreign body giant cells. It was also found that DC-STAMP is an essential cell-cell fusion regulator in macrophage lineage cells.
