【 要旨 】 PTHrPはおもに，パラクラインあるいはオートクラインの様式で，局所で分泌され局所で作用する．この点で内分泌ホルモンであるPTHとは大きく異なる．ノックアウトマウスの表現型から，生理的に成長板軟骨細胞の分化を制御していることが明らかにされた．その後，組織特異的にPTHrPを過剰発現させたトランスジェニックマウスや軟骨細胞特異的にPTHrPシグナルを戻すことで致死性を回避したレスキューマウスの作出によって，ほかの組織でも重要な役割を果たすことが示された．本稿ではおもに最近の成果に重点を置き，その多彩な生理作用について述べる．

[ Summary ] PTHrP functions mainly locally as a paracrine/autocrine factor, while PTH works as an endocrine hormone. Studies of PTHrP-/- mice revealed that PTHrP regulates differentiation of growth plate chondrocytes. Subsequent studies of transgenic mice overexpressing PTHrP in cartilage and rescued PTHrP-knockout mice showed that it also plays an important role in other tissues. This article focuses on recently discovered variable physiological functions of PTHrP.
