Vitamin D negatively regulates parathyroid hormone (PTH) synthesis and secretion as well as parathyroid cell growth in patients with chronic renal failure. This feedback action of the active vitamin D metabolites mediates not only through vitamin D receptor as genetic action, but also via serum calcium level, which is another strong regulator through calcium-sensing receptor in parathyroid cells. Here, we review the molecular mechanism and action of vitamin D, including newly developed vitamin D analogs, from the therapeutic view point of renal hyperparathyroidism. |
