【 要旨 】 腹膜透析(PD)の限外濾過にaquaporin(AQP)1の関与が指摘されている.しかし,その発現調節機構については明らかにされていない.従来AQP 1はハウスキーピングタイプで構成的に細胞膜上に存在するといわれてきたが,われわれは腹膜中皮細胞におけるAQP 1のshort-term regulationの可能性について検討した.高浸透圧培養液(5%グルコース,マン二トール)に室温で15分間浮遊させるとラット培養腹膜中皮細胞の細胞膜上AQP蛋白量は通常の培養液群に比べ有意に増加していた.この増加は2.5分間から観察された.これより腹膜中皮細胞のAQP 1は高浸透圧刺激で細胞内から細胞膜上へトランスロケーションする可能性が示唆された.PDの限外濾過には,AQP 1トランスロケーションの関与があると考えられる.

[ Summary ] Aquaporin-1 (AQP 1) has been reported to play an important role in ultrafiltration through the peritoneum in peritoneal dialysis. To determine the precise mechanism involved in this process, the present study was designed to examine the translocation of AQP 1 to the plasma membrane, induced by hyperosmotic stimuli. Cultured RPMC obtained were incubated in a medium with or without glucose or mannitol as the hyperosmotic stimuli, at room temperature.
Significant enhancement of AQP 1 quantity by the addition of 5% glucose to medium was observed at 2.5 minutes, followed by a sustained increase in quantity over the next fifteen minutes. AQP 1 quantities were also significantly increased by the addition of 5% mannitol. These results suggest that hyperosmotic stimuli could generate an augmentation of AQP 1 quantity in the plasma membrane by the translocation of AQP 1.
This process, in the short term, might contribute to the mechanism involved in ultrafiltration through the peritoneum in peritoneal dialysis.
