【 要旨 】 一酸化窒素(NO)は胃粘膜統合性の維持において他の調節因子とともに重要な役割を担っており,種々の傷害惹起物質から胃粘膜を防御するのみならず,損傷の修復機転においても促進的な影響を及ぼしている.しかし,他の臓器と同様に胃粘膜においてもNOの二面性が指摘されており,ある種の胃疾患では病因物質としてのNOの作用も認められている.一般的に構成型NO合成酵素(cNOS)および誘導型NOS(iNOS)由来のNOはそれぞれ粘膜保護的および傷害性に作用するものと考えられているが,最近,iNOS由来のNOも保護的に作用する事例が示されており,NOの二面性の詳細については今後の検討課題となっている.

Title Roles of Nitric Oxide in Gastric Mucosal Damage Protective and Proulcerogenic Actions

[ Summary ] Nitric oxide (NO) is synthesized from L-arginine by an enzyme, called NO synthase (NOS). Two types of NOS have been identified ; one is a constitutive NOS (cNOS), that is expressed under normal conditions, and the other referred to as inducible NOS (iNOS), is induced by inflammatory cytokines. The NO produced by CNOS acts as a transduction molecule underlying various physiological responses, whereas that produced by iNOS acts as a cytotoxic molecule to kill the invade microorganism or tumor cells. The dual actions of NO are also seen in the stomach : cNOS/NO plays a role in maintaining mucosal integrity, in collaboration with other protective factors such as prostaglandins(PGs) and sensory neurons. However, iNOS/NO exerts complex influences, playing the part of both a pathogenic and a protective molecule, depending on the particular experimental conditions. Further studies should be done to disclose the interactive role of NO in relation to other protective factors, such as PGs, as well as the nature of the dual reactions induced by NO in the gastric mucosa.
