【 要旨 】 潰瘍性大腸炎(UC)の炎症粘膜では加齢により認められるDNAメチル化(agerelated methylation)の亢進が顕著であり,腫瘍合併例でその頻度,割合が高いことよりDNAメチル化がUCにおける炎症性発癌の早期の重要なメカニズムであると考えられる.DNAメチル化異常は前癌病変においても検出できるという点よりバイオマーカーとして応用できる可能性が高く,メチル化される遺伝子群の特定のパスウェイ同定は分子標的治療や化学予防にも繫がることが期待される.

[ Summary ] DNA methylation has been deeply involved in the development and progression of digestive system cancer. Aberrant DNA methylation has also often been observed in aged and inflammatory digestive tissues such as ulcerative colitis (UC). Accelerated age-related DNA methylation has been observed in the colonic mucosa in UC patients with neoplasia, suggesting that DNA methylation would be an important initial mechanism of UC-related tumorigenesis. Since DNA methylation can be detected in precancerous tissue, it could potentially be useful as a marker for risk estimation of cancer development. Moreover, identifying specific pathways of methylated genes in UC-related tumorigenesis raises the possibility that specific epigenetic therapies could be useful in the treatment or chemoprevention of UCrelated colorectal cancer.
