【 要旨 】 ヒト消化器癌は慢性炎症組織を母地に発生するものが多く,慢性炎症の制御が炎症性発癌の抑制に重要であることが知られている.従来,詳細な炎症性発癌メカニズムには不明な点が多いとされてきたが,近年になり慢性炎症が遺伝子異常やエピジェネティクス変化を引き起こす分子機構が徐々に明らかになってきている.将来的に,炎症性発癌の分子メカニズムの解明が癌の予防および治療につながることが期待される.

[ Summary ] Many types of digestive organ cancers, such as hepatocellular carcinoma, gastric cancer, colitis-associated colon cancer, bile duct cancer and esophageal cancer from Barrett's esophagus, arise from tissues with chronic inflammation. Controlling chronic inflammation is important for inhibiting cancer development ; however, the detailed mechanisms of inflammation-associated carcinogenesis remain unclear. Recent progress in molecular biological research has revealed how chronic inflammation can induce genetic abnormalities. It is becoming increasingly obvious in both basic research and epidemiological studies that these genetic changes are closely associated with cancer development. In addition to genetic aberrations, epigenetic modifications in cancer tissues are also implicated in cancer progression. Elucidation of the mechanisms of inflammation-associated carcinogenesis promises to be useful for cancer prevention and treatment in clinical practice.
