[ Summary ] Chronic inflammation plays an important role in the development of ulcerative colitis (UC) associated colorectal cancer (UCAC). Accelerated age-related DNA methylation has been observed in the colonic mucosa in UC patients with neoplasia, suggesting that DNA methylation may be an important initial mechanism of UC-related tumorigenesis. On the other hand, somatic mutations in the tumorsuppressor gene TP53 are frequently observed in UCAC in early stage disease. More detailed interactive mechanisms of age-related DNA methylation and TP53 mutation in UC-related tumorigenesis remain to be clarified.
